Ipamorelin is a selective pentapeptide GHRP and ghrelin receptor agonist. Its selectivity for GHS-R1a distinguishes it from earlier GHRPs — it stimulates GH release without meaningfully activating ACTH, cortisol, or prolactin pathways.
Binding to GHS-R1a in the hypothalamus and anterior pituitary triggers GH release through phospholipase C and PKC signaling. The pulsatile GH release ipamorelin produces mirrors physiological GH secretion patterns. IGF-1 levels rise downstream, promoting anabolic and regenerative signaling.
Studies show 2–3× increases in GH pulse amplitude without cortisol elevation (vs GHRP-6 which raises cortisol ~80%). Animal studies document improved bone mineral density, increased lean mass, reduced fat mass, and accelerated wound healing with prolonged administration.
