GHRP-6 is one of the original synthetic hexapeptide GH secretagogues, functioning as a ghrelin receptor agonist. It produces strong GH release and is notable for its significant appetite-stimulating effects — a direct consequence of ghrelin receptor activation in the hypothalamus.
GHRP-6 ghrelin receptor agonism in the hypothalamus strongly activates NPY/AgRP hunger neurons, producing marked increases in appetite. GHRP-6 also modestly elevates cortisol and prolactin compared to ipamorelin. This profile has made it a key tool for studying the orexigenic arm of the ghrelin system.
GHRP-6 was among the first GH secretagogues in clinical research, with data spanning 30+ years. Studies established dose-response relationships for GH stimulation, characterized appetite effects, and demonstrated cardiac protection in ischemia models via a GH-independent mechanism.
