5-AMINO-1MQ

5-Amino-1MQ is a small molecule inhibitor of NNMT (Nicotinamide N-Methyltransferase), an enzyme that methylates nicotinamide using SAM as the methyl donor. NNMT is highly expressed in white adipose tissue and inhibiting it raises intracellular NAD+ and SAM levels.

NNMT inhibition in adipocytes reduces fat cell size, promotes adipose tissue browning (conversion of white fat to metabolically active beige fat), raises NAD+ for sirtuin activation, and improves insulin sensitivity. 5-Amino-1MQ is structurally optimized for NNMT selectivity and cellular penetration.

Published studies in obese mouse models demonstrate: 7% body weight reduction, significant reduction in adipocyte size, upregulation of brown fat markers (UCP-1, PGC-1α), improved glucose tolerance, and elevated plasma NAD+ levels. The mechanism is distinct from all GLP-1 and GH-based approaches.
